Melina Kanakaredes was born on April 23, 1967, in Akron, Ohio to an insurance salesman Harry Kanakaredes and his wife Connie (née Temo), a candy company owner. Melina is a second-generation Greek-American and speaks Greek. She has two sisters Lia Kanakaredes and Aretta Kanakaredes.

She became passionate about acting at a very young age and by the time she turned 8, she made her stage debut in a production of Tom Sawyer at Weathervane Playhouse in Akron, Ohio. She attended the Firestone High School in Akron and after her graduation, she moved to Ohio State University for a short time, and then transferred to Point Park College in Pittsburgh, Pennsylvania. She left the show in 1995 and landed the role of Victoria Metcalf in “Due South” in the same year.
